Increased penetration depth
Using confocal microscopy to image organoids, especially those larger than 120 micrometers, presents challenges due to limited penetration depth, making it difficult to capture internal structures. The high laser intensity required for deeper imaging can cause phototoxicity and photobleaching, damaging cells and reducing image quality. Additionally, extended acquisition times may disrupt dynamic cellular processes.
